Non-thermal plasma has been shown to be very promising for different biomedical applications. This is due to the fact that plasma generates reactive oxygen and nitrogen species that can have both regenerative and cytotoxic effects according to their doses. Hydrogels have been studied as potential media for the storage and delivery of plasma-generated RONS due to their capacity to remain in the site of interest and release RONS in a controlled way. Furthermore, hydrogels have been studied as model systems to evaluate the effect of direct plasma treatment in living tissues. The aim of this work is to study the distribution of RONS on the surface of gelatin and agarose hydrogels and the penetration depth through them. Moreover, a quantification of the transport of RONS through thin hydrogel layers in a water solution is studied. We are comparing the effect of plasma treatment on gelatin and agarose within the same study. Gelatin and agarose hydrogels were treated by an atmospheric pressure plasma jet using helium. Two different kinds of experiments were performed. In one of them, the hydrogels contained colorimetric probes to evaluate the distribution and penetration depths. The change of colour on the surface and inside the hydrogels was evaluated qualitatively, by a visual analysis, and semi-quantitatively, with the software ImageJ. The results showed that RONS distribution increased with the treatment time and it had a radial distribution on the hydrogel surface. RONS concentrations were higher in agarose than in gelatin hydrogels. For the penetration depths, similar results have been obtained. For the second experiment, we evaluated the RONS concentration in a water solution putting a hydrogel layer between them. The quantification was done using chemical probes and showed a linear dependence of tRONS concentration with the treatment time, and an inverse dependence with the hydrogel thickness. Moreover, it is also observed a higher RONS concentration in agarose.

Diabetes mellitus is a metabolic disease characterized by defects in insulin action or secretion, resulting in chronic hyperglycemia. One of the most common complications of this condition is the development of chronic wounds, particularly diabetic foot ulcers, which can lead to infections, and amputations. Current treatments for these wounds, such as antibiotics, are facing alarming challenges mainly due to the increasing antibiotic resistance crisis. In this context, Cold Atmospheric Plasma (CAP) technology has emerged as a potential solution for infection treatments. This technology generates reactive oxygen and nitrogen species (RONS) that selectively kill bacteria without harming healthy cells. Furthermore, indirect treatment methods using plasma-treated liquids have been developed. Thereby, using these liquids as base for the formation of hydrogels could offer promising opportunities in many biomedical applications. Additionally, the incorporation of hyaluronic acid (HA) in hydrogels can enhance tissue regeneration processes. This biomolecule, present in the extracellular matrix, promotes wound healing by inflammation modulation, cellular migration and angiogenesis. Therefore, this project aims to combine these strategies by using hydrogels to these diabetic chronic wounds. The incorporation of RONS derived from cold plasma in hydrogels targets bacterial infections, while the inclusion of HA promotes tissue regeneration. With the experimental work done in the present study, it was seen that RONS can be successfully transferred to hydrogels, giving them an antibacterial response. They show killing rates of over 95% for treatments of 20 minutes. Also, HA does not affect these antibacterial effects, so it can also be implemented.

Plasma medicine has experienced an international boom in recent years. Plasma, considered the fourth state of matter, is defined as an ionized gas composed of free electrons, ions, free radicals, electromagnetic fields and reactive species, among other substances. Recent studies have demonstrated the medical applications of reactive oxygen and nitrogen species (RONS) generated in cold plasmas at atmospheric pressure (CAP). Numerous investigations directly link RONS to the modification of oxidative stress in cells, thus opening the possibility of their application in cancer therapies. The present study focuses on indirect treatments using CAP. These treatments consist of the use of a plasmatreated biocompatible liquid or hydrogel that is used as a vehicle to deliver RONS to the specific site affected by the tumor. Currently, the research group, with which the present study collaborates, has patented three different technologies for indirect treatments: a plasma-treated liquid (PTL), a plasma-treated hydrogel (PTH) and a composite from a plasma-treated hydrogel and calcium phosphate (PTH Composite). The latter treatment specializes in treating osteosarcoma, the most common bone cancer, by promoting bone regeneration and anti-cancer treatment simultaneously. This Final Degree Project pursues two main objectives, on the one hand, to perform the qualification and classification for each of the technologies of the group (PTL, PTH and PTH Composite), taking into account the current regulatory standards in Europe and the different nature of each product. In addition, once the three technologies have been classified and qualified, the first steps necessary for their commercialization will be identified. The other main objective of the work is the physicochemical characterization of the plasma-treated liquid (PTL). Specifically, the purpose is to be able to provide information on the stability of RONS in a PTL, a fundamental aspect for the development of the technology. According to the results obtained, the stability of the PTL is directly related to the pH of the liquid. By controlling the acidity of the medium, it has been possible to extend the shelf life of the PTL, thus modifying its stability. These results should be taken into account when designing and characterizing new PTL/PTH in the future.

In recent years, Plasma Medicine has become a field to study with great potential due the results that it has shown in some studies realized around the world. Plasma is defined as an ionized gas that is generated by applying an electrical discharge to a gas, helium (He) in this case, and which contains, among others, numerous reactive oxygen and nitrogen species (RONS). These species induce an increase in oxidative stress in cells that is being studied as a new treatment against cancer, less invasive and highly selective, which avoids damaging the patient's healthy cells. This Final Degree Project focuses on the optimization of alginate hydrogels as vehicles for the release of these reactive species generated by plasma. This is carried out by studying what parameters affect the release of species using a non-thermal plasma treatment. The aim for this work consists in use these hydrogels to produce implants that maintain a concentration of these species longer than other liquid vehicles at the site of the tumor. All the materials used in this work are biocompatible, thus avoiding complications due to degradation or reactions of the materials with body fluids. The reactive species generated by non-thermal plasma are very varied, and in this work, we will focus on two in particular, H2O2 and NO2 - that we generate with the Helium plasma discharge with 0.3% synthetic air that allows the formation of these species, among others. To analyze the RONS delivery, some release or degradation tests are carried out, thus being able to verify the behavior of this biomaterial and compare the results with the biopolymer itself. During the work it has been possible to evaluate different methods of preparation and reticulation of the hydrogels, achieving an expected release of these species in a period of time of 70 minutes in a medium with conditions similar to the physiological ones, in addition it was possible to analyze the influence of the microstructure and the rheological properties of the hydrogel in the release process.

En els darrers anys, la tecnologia de plasma no tèrmic ha sorgit amb un gran potencial en aplicacions en el camp biomèdic. El plasma està compost per una gran varietat d'espècies reactives, entre les quals trobem espècies reactives d'oxigen i nitrogen (RONS), que poden actuar sobre una mostra determinada. Entre les aplicacions mèdiques d'aquesta tecnologia podem trobar l'ús d'aquestes espècies reactives derivades del plasma en tractaments contra el càncer amb l'objectiu de desenvolupar procediments menys invasius i menys agressius ja que s'evita danyar les cèl·lules sanes. Aquest treball, es centra en com hidrogels compòsits basats en gelatina influeixen en l'estabilitat i l'alliberament d'espècies reactives generades en solucions de biopolímers mitjançant el tractament amb plasma no tèrmic. Aquestes solucions biopolimèriques compostes per Dermatan i Àcid Hialurònic, seran tractades per augmentar la quantitat de RONS en comparació amb els líquids tractats amb plasma. L'ús d'hidrogels de gelatina es justifica per crear implants que siguin fàcils de manipular i per evitar que la solució de biopolímers sigui arrossegada pels fluids corporals després d'implantar-los. Tots els components amb els que es treballarà son d'origen biològic, biocompatibles i biodegradables per evitar interaccions no desitjades amb el cos humà. La investigació de la capacitat d'emmagatzematge de les solucions de biopolímers es va centrar en el H2O2, NO2 - i NO3 - , els quals seran generats amb un plasma no tèrmic alimentat amb una barreja d'He + 0.3 % d'aire sintètic. A més, per avaluar el comportament dels hidrogels de gelatina, es van fer assajos de degradació. Després es va avaluar la generació i l'estabilitat dels RONS produïts. Els resultats es van comparar en funció del biopolímer utilitzat per emmagatzemar RONS juntament amb els mètodes de detecció emprats. Durant els experiments, es va observar un augment dels RONS generats a la solució de Dermatan i àcid hialurònic, en comparació amb els generats en PBS. Vam poder avançar un potencial alliberament continuat dels RONS dels hidrogels després de 45 minuts en condicions fisiològiques.

Non-thermal plasma technology has been largely studied in the last decades for many applications, and medicine is among the most promising ones. Working at room temperature allows to treat human tissues and to develop medical applications among them the cancer therapy. Non-thermal plasmas can generate reactive oxygen and nitrogen species that have the ability to interact with biological systems. In particular, they are selective in killing cancer cells without damaging the healthy cells present in the surrounding tissues. Internal cancers require to use indirect treatments and plasma treated liquids. However, the injection of these liquids directly in human body would not be beneficial since the reactive species contained in them would be quickly washed away by the body fluids. Generation and storage of reactive species in suitable delivery systems like hydrogels, could offer a promising and viable way to treat cancers. Indeed, the hydrogels can be implanted by surgery or be injected before crosslinking and release locally the reactive species which can interact effectively with tumours. In this project, solutions of heparin, chondroitin and albumin were studied as possible media for reactive species generation by plasma treatment and storage. Three concentrations of each polymer (1%w/w, 5%w/w, 10%w/w for heparin and chondroitin and 0.5%w/w, 1%w/w, 2.5%w/w for albumin) were plasma treated to assess the influence of polymers on the production of reactive species and to select the optimal concentration for each polymer. Hydrogels were produced by mixing polymers with agarose 4% w/w. The mechanical properties and the microstructure of the hydrogels were studied by rheology and scanning electron microscopy, respectively. Finally, the release properties of hydrogels loaded with plasma-generated reactive species were studied in phosphate buffer saline at 37 °C.

Osteosarcoma is a type of cancer that mainly affects children and young people, and has an aggressive treatment usually based on a combination of surgery and chemotherapy with relatively low survival rates. This project aims to optimize and characterize a composite biomaterial made of calcium-deficient hydroxyapatite microspheres in a hydrogel matrix of alginate and gelatin. The hydrogel, through a cold plasma treatment at atmospheric pressure, can generate reactive oxygen and nitrogen species, which have been related to an increase in preferential oxidative stress in tumor cells, inducing the cell death of cancer cells without affecting the healthy cells. In addition, this material has the potential to regenerate bone thanks to the incorporation of a bioceramic phase in it, forming a composite biomaterial. For this, the synthesis of the material has been optimized to ensure its homogeneity. In addition, some physical characteristics have been studied, such as the viscosity and injectability of the hydrogel, and the influence that both microspheres and plasma treatment have on them. In the same way, the release of reactive species generated by plasma into an aqueous medium and in a medium with a calcium concentration similar to that present in the possible area of application has been evaluated. On the other hand, the effect of the size of hydroxyapatite microspheres deficient in calcium on the physical properties of the composite material and its effect on the release of reactive species has been studied.

Non-thermal plasmas are ionized gases at room temperature composed of free electrons, ions, radicals, excited atoms and molecules, neutral molecules and electromagnetic radiation. They are highly reactive systems that are exploited in multiple fields, ranging from material production and modification, environment, agriculture and food processing and medicine. This high versatility is mainly due to the ability to control the type and amount of reactive species (RS) produced during a plasma treatment. The primary RS, generated in a non-thermal plasma, react rapidly with the gas molecules to generate secondary RS that are less reactive and therefore can live longer. If the plasma is generated close to a liquid, the secondary RS can diffuse in the liquid and react with molecules dissolved in it. The nature of the gas and the liquid determine which RS are generated and their amount. The aim of this project is to study the influence of the feed-gas on the RS generated in water solution by an atmospheric pressure plasma jet. The gases that will be investigated will be pure helium and helium doped with molecular oxygen, nitrogen, or water vapour. The presence of RS in the gas-phase will be investigated by optical emission spectroscopy while the amount of RS in water will be quantified by colorimetric and/or fluorescent chemical probes.

Recently, the application of cold atmospheric plasma has been shown to be very promising for the growth of applications in the area of biomedicine. This is due to the fact that a plasma is composed of a great variety of active elements, such as electromagnetic fields, UV-vis-IR radiation and reactive chemical species that act synergistically on a treated sample. One of the most promising application of plasma in the biomedical sector is their use in dermatological treatments. This project studies the physico-chemical effects induced by the application of plasma to gelatine solutions as a model of the skin for dermatological treatments. The project also investigates the possible application of plasma for tattoo removal since plasma is known to degrade organic dyes in solution. The use of gelatine as a skin model is due to the fact it is the main proteic component of skin, in the form of collagen. To achieve these objectives, gelatin samples were treated by an atmospheric pressure plasma jet using two different modes of application, point treatment and surface treatment. The effects of the treatments on the hydrogel were analysed by means of rheology techniques and Raman spectroscopy. We studied also in details the temperature effects and the evaporation induced by the plasma treatment on the samples. In addition, to determine the effects of plasma on tattoo inks in liquid and solid solutions, optical measurements were performed before and after the treatment. The techniques used to carry out these experiments were UV-Vis absorbance spectroscopy and colorimetry. The results were carefully evaluated and compared with other existing methods for tattoo inks removal.

The early detection of eye disease is essential to be able to act and avoid the loss of vision. Part of these diseases are related to the loss of the photoreceptors of the retina. For that reason, that the visualisation of all the elements of the retina is particularly important. Conventional technologies do not offer enough resolution to visualize the photoreceptors. Here is where importance of the study in new microscopy techniques, such as AOSLO, relies. Additionally, the characterization and positioning of those is the clue to make a reliable diagnosis. The present project has been focused on the development of the support programs used to validate the new microscopy technics, AOSLO, as a help to diagnose. These techniques are characterised by adaptative optics set able to improve the resolution of the images which are already being taken in conventional ophthalmology clinics. As a new technique, specialists responsible for the diagnoses need tools to interpretate the images. In that point, the developed program helps to transform the adaptative optics images into distance between cones, a biological marker. This step begins introducing functions to the program that develop an image where there can be done a Fourier transform. The Fourier transform is characterized for the values that this project has been optimizing. As a result, there is the obtention of images and graphs that apport values for an objective diagnose.

En els últims anys ha cobrat importància l'ús dels plasmes atmosfèrics freds en aplicacions biomèdiques, ja que que ofereixen la possibilitat de tractar teixits vius per la seva baixa temperatura de tractament, i les espècies reactives que produeixen a l'tractar líquids permeten una acció terapèutica similar en teixits vius. Investigacions recents han demostrat que el tractament amb plasma té diverses aplicacions d'interès (esterilització de dispositius mèdics, curació de ferides, tractament de cancer) El grup de Medicina Plasma del BBT (UPC) utilitza aquesta tecnologia per estudiar un possible tractament per l'osteosarcoma, en combinació amb hidrogels de biopolímers dins el projecte APACHE (Atmospheric Pressure plasma meets biomaterials for bone Cancer Healing) finançat pel Consell Europeu de Recerca (ERC). Els biopolímers estan emergint com materials adequats per a moltes aplicacions en medicina, per la seva versatilitat i biocompatibilitat. El tractament amb plasma d'hidrogels és molt novedos, i s'ha vist que pot produir una gran quantitat d'espècies reactives d'oxigen i nitrogen que poden emmagatzemar-se de manera efectiva i alliberar-se a l'entorn biològic. S'ha demostrat que aquestes espècies maten selectivament les cèl·lules canceroses si es dosifiquen adequadament, però es desconeix encara com es modifica el polímer per efecte de l'tractament de plasma i quines repercussions pot tenir això. L'objectiu d'aquest projecte de grau final és doble: d'una banda explorar la literatura per tal de trobar possibles estratègies per abordar l'efecte de l'tractament amb plasma fred a les solucions d'alginat a nivell molecular que depenen de l'ús de molècules traçadores; descobrir com altres grups han emprat molècules traçadores per estudiar els efectes dels plasmes freds en un sistema més complex i identificar les característiques d'una bona molècula traçadora. D'altra banda, en la segona part del projecte, apliquem aquest coneixement per a dissenyar i validar un possible model per alginat. A la primera part, recopilem diversos estudis, centrats en avaluar els efectes de l'plasma fred a les molècules traçadores més rellevants (fenol, cisteïna i glutatió). A l'identificar l'oxidació d'aquestes molècules és possible extrapolar els resultats a sistemes més complexos i trobar correlacions entre les fonts i paràmetres del plasma, la producció de RONS i els resultats biològics. A la segona part, identifiquem el glucoronat com a possible model per l'alginat i l'investiguem experimentalment en termes de producció d'espècies reactives mitjançant tractament amb plasma. Quantifiquem les espècies reactives (peròxid d'hidrogen, ions nitrit i radicals OH) produïdes en solucions d'alginat i glucoronat utilitzant sondes químiques colorimètriques i fluorescents i les comparem amb la quantitat produïda en el tampó fosfat. Vam descobrir que la quantitat d'espècies reactives produïdes en solucions d'alginat i glucoronat amb la mateixa concentració és la mateixa, el que significa que el glucoronat és un bon model a nivell molecular. A més, confirmem que la quantitat de peròxid d'hidrogen i nitrits produïts és més gran que en el tampó fosfat, mentre que la quantitat de radicals hidroxil és menor, el que significa que alguns d'ells estan reaccionant amb la matèria orgànica present en la solució. També vam començar alguns experiments preliminars per avaluar l'efecte de l'tractament amb plasma sobre solucions d'alginat i glucoronat mitjançant espectroscòpia infraroja i per mapejar la generació i difusió d'espècies reactives en solucions usant una sonda colorimètrica.

The use of cold atmospheric plasma in cancer treatments has been shown to be effective in creating a selectivity between healthy cells and cancer cells as a result of the contribution of reactive oxygen and nitrogen species (RONS) to the cells. Complementing it with an anti-cancer drug commonly used in current treatments, such as doxorubicin, greater effectiveness could be achieved as well as a reduction of the amount of antitumoral drug used in such therapy. In the present project, it is intended to synthesize and characterize a material composed of a hydrogel treated with cold atmospheric plasma and calcium-deficient hydroxyapatite microspheres loaded with doxorubicin, for its posterior use as dual release platform for bone cancer therapy. For this, microspheres have been successfully synthesized by emulsion of a calcium phosphate cement and microspheres have been loaded with doxorubicin. The microspheres have been added to a hydrogel composed of gelatin and alginate previously treated with cold atmospheric pressure plasma. The release of RONS and doxorubicin has been studied from the designed material to an aqueous medium. In parallel, it has been evaluated the relevance of proceeding to the coating of microspheres with polycaprolactone (PCL) by plasma polymerization and it has been studied how it influences the release kinetics of doxorubicin from the composite material. The results indicate that the composite material releases RONS and doxorubicin to the release medium.

Els ossos sans es troben en constant un remodelatge que fan ells mateixos per tal de satisfer les necessitats de cada individu. Algunes malalties com l'osteoporòsi, els osteosarcomes o trencaments generen una inestabilitat en la remodelació òssia i no poden garantir-la per tant, es necessita un implant. Les ceràmiques de fosfats de calci (CaP) són la millor opció com a implants sintètics per la seva composició que imita la de l'os. Els CaP també son reabsorbibles i no toxics, de manera que no hi ha necessitat d'una segona cirugia ja que l'area ocupada per l'scaffold anirà gradualment sent substituida per nou os. Degut a les seves propietats, el cold atmospheric plasma forma part d'estudis oncològics. La seva capacitat d'eliminar cèl·lules cancerígenes sense causar efecte en la resta del teixit gràcies a la seva habilitat per generar espècies reactives (RONS) que són necessàries pels processos biològics converteixen al plasma en una opció amb molt potencial. En els grups de recerca del BBT s'esta estudiant una estratègia nova que consisteix en transmetre aquests RONS a un hidrogel per tal de portar aquests RONS a la zona del afectada. Conservant així tant els beneficis del plasma com les popietats de l'hidrogel. Aquest hidrogel es podrà combinar amb el CaP per tal que l'implant final tingui els avantatges tant del plasma com dels fosfats de calci per a la regeneració òssia. El propòsit d'aquest estudi és investigar mitjançant un estudi in vivo les propietats biològiques i la seguretat dels scaffolds de fosfats de calci per regeneració òssia i el potencial de l'efecte dels tractaments de plasma sobre aquests. Després de dos mesos d'implantació en un defecte creat al condil femoral de conills, els resultats obtinguts amb SEM, µCT i microscopi òptic demostren que l'os ha crescut a la superfície de l'implant, demostrant la bona adhesió sense cap resposta inflamatòria. Després d'analitzar les imatges mitjançant softwares com el cTAN I l'ImageJ, s'ha calculat el percentatge d'os nou regenerat de cada tipus de mostra. Es conclou que el millor tipus de mostra és la formada per l'scaffold de CaP en combinació amb l'hidrogel que ha estat tractat amb plasma.

Bone is a hard tissue which is remodelled throughout life and possesses the ability to heal small injuries and ruptures, meanwhile larger fractures, or voids generated after a tumour removal, are more difficult or even impossible for the bone to heal by itself. Thus, bone substitutes are required and natural bone substitutes from the patient itself, from a human cadaver donor or from an animal are nowadays current options for bone repair. The main problem when using natural bone substitutes is the risk for disease transmission or rejection, the limited amount of the bone substitute in autografts and some ethical considerations. For these reasons, new synthetic bone substitutes have been developed the past years. One class among the many biomaterials applied for synthetic bone substitution are Calcium Phosphate Cements (CPCs). This report focuses on the development of Calcium Phosphate Cement (CPC) microspheresfrom alpha Tricalcium Phosphate (α-TCP) as synthetic bone substitution and drug carrier of the chemotherapeutic drug doxorubicin. Once the microspheres synthesized and loaded with the antitumoral drug, the main goal is to study the impact on the doxorubicin release kinetics from polymer-coated microspheres to a physiological receptor medium (PBS). In order to avoid loss of the incorporated doxorubicin by using a chemical wet process, polymer coating of the microspheres was carried out by low-pressure plasma polymerization. Leaning on their biocompatible properties, poly(ethylene glycol) (PEG) and poly(εcaprolactone) (PCL) layers were selected for coating the doxorubicin-loaded microspheres, using the monomers Diethylene Glycol Dimethyl Ether (DIGLYME) and ε-Caprolactone (ε-CL), respectively, as precursors for plasma polymerization. By performing a sieving of the microspheres after their synthesis, the doxorubicin loading capacity was evaluated for microspheres with different size distribution and it was revealed that microspheres of the size 100 < ∅ < 150 μm have a greater loading capacity, 60 % of the maximal amount available, meanwhile microspheres of the size 250 < ∅ < 300 μm have a lower loading capacity, 50 % of the maximal amount available, which can be related to their respective specific surface area (SSA). It was also found that the release kinetics of doxorubicin to Phosphate-Buffered Saline (PBS) is sizeindependent, although smaller microspheres causes a greater amount of doxorubicin to be entrapped within the bulk material. Hence, further investigations regarding long-term cytotoxicity are required. During this project, in order to study the relevance of the use of plasma polymerization in the design of the final doxorubicin microsphere carriers, the effects of different monomers, plasma polymerization times as well as single or double polymerization steps of the microspheres were evaluated on the doxorubicin release. It was proven that poly(ethylene glycol)-coatings, using Diethylene Glycol Dimethyl Ether (DIGLYME) as precursor, do not form thick enough polymer-coatings to completely cover the microspheres and therefore is not resulting in any specific impact regarding the doxorubicin release. Hence, it was decided to continue the experiments using only poly(εcaprolactone)-coatings. Indeed, poly(ε-caprolactone)-coatings revealed a decrease of the doxorubicin release into Phosphate-Buffered Saline (PBS) from the microspheres, when performing release kinetics for eight hours. It was found that, when the specific surface area is lower, which is the case when working with microspheres of the size 150 < ∅ < 300 μm, increasing the plasma polymerization time is enough to observe an influence on the doxorubicin release and therefore the effect from the doublelayered poly(ε-caprolactone)-coating is not shown nor required in this case. On the other hand, when the specific surface area is higher, which is the case when working with microspheres of the size 100 < ∅< 150 μm, a double-layered poly(ε-caprolactone)-coating is required in order to decrease the amount of doxorubicin released. In the best of our knowledge, this report presents for the first-time calcium phosphate cement microspheres successfully polymerized at room temperature by using low-pressure plasma polymerization process. According to the observations made, the poly(ε-caprolactone)-coated microspheres show promising results regarding doxorubicin release.

Calcium phosphate microspheres/microparticles will be synthetized in order to be included into a hydrogel matrix with the aim to design hydrogel-based biomaterials for bone cancer therapy in which the calcium phosphate microparticles will be reservoirs of drugs of oncological interest. While stability and optimization of the system will be studied by physic-chemical experimental techniques, drug loading of the calcium phosphate microparticles and its subsequent release from the designed biomaterials will be evaluated by drug release experiments.

En los últimos años, la medicina de plasma ha sido un tema emergente de estudio e investigación en distintas áreas (esterilización, curación de heridas, tratamientos dentales), y muy especialmente en el área de terapias de cáncer. Estudios recientes han demostrado que las especies reactivas de oxígeno y nitrógeno que genera el plasma tienen un efecto selectivo y citotóxico en las células cancerígenas, sin afectar a las células sanas. Se han demostrado efectos letales del plasma en más de 20 líneas celulares in vitro e in vivo, y recientemente, en el grupo de investigación BBT, en cáncer de hueso. El tratamiento plasma puede efectuarse de forma directa, aplicando directamente el plasma sobre el tumor o las células, de modo que todos los componentes del plasma interaccionan con el material biológico; o de forma indirecta, poniendo en contacto con el tumor o las células un medio líquido previamente tratado con plasma, de manera que únicamente las especies reactivas que se generan en el medio líquido interaccionan con el material biológico. El objetivo principal de este proyecto es investigar los efectos directos e indirectos del plasma a presión atmosférica sobre dos líneas celulares de osteosarcoma, y estudiar dichos efectos tanto en 2D (células sembradas en placas de cultivo) como en 3D, haciendo crecer las células tumorales en andamios tridimensionales para mimetizar mejor el microambiente tumoral. Para ello, se han caracterizado las especies reactivas (H2O2, NO2-, y especies de tiempo de vida corto) generadas por el plasma atmosférico en medio de cultivo a diferentes tiempos de tratamiento. La concentración de especies en el medio aumenta progresivamente con el tiempo de tratamiento. Para realizar el estudio del efecto del plasma en 3D, se han sintetizado nanopartículas de hidroxiapatita para incorporarlas a andamios de colágeno. Se ha analizado la degradación y capacidad de absorción de líquidos de los andamios con el fin de comprobar su aptitud para la siembra de células. Posteriormente se ha estudiado los efectos directos e indirectos del plasma sobre las células sembradas en los andamios. Se ha comprobado que la concentración de especies reactivas que se generan es proporcional al tiempo de tratamiento, y en contraste con trabajos anteriores, se ha observado que la viabilidad de las células tratadas por plasma de manera indirecta no se ve afectada. Esto es debido a que se ha empleado medio de cultivo con piruvato, que actúa como scavenger de los peróxidos. Con el tratamiento directo sí que hay un ligero efecto citotóxico en las células, tanto en 2D como en 3D.

Els stents biodegradables o reabsorbibles són la nova generació d'stents fabricats amb biomaterials biodegradables recoberts amb els fàrmacs antiproliferatius d'alliberament controlat i que tenen l'avantatge que desapareixen al cap d'uns mesos. D'aquesta manera és possible la recuperació de la funció arterial, cosa que no succeeix amb els stents metàl·lics o els stents alliberadors de fàrmac. L'objectiu del present treball és fabricar stents de poli-L-làctic àcid (PLLA) carregats amb àcid acetil salicílic (AAS) amb la tècnica d'impressió en 3D i caracteritzar-los de forma físico-química i biològica amb cèl·lules endotelials (EC) així com determinar el temps d'alliberament del fàrmac. També s'han fabricat films de PLLA amb aspirina per la tècnica de dissolució i colada per tal de facilitar tècniques d'anàlisi com l'angle de contacte (CA) o la calorimetria diferencial d'escombratge (DSC). Els resultats obtinguts mitjançant la tècnica de l'angle de contacte permeten relacionar l'aplicació d'un tractament tèrmic sobre els films amb una degradació de l'àcid acetil salicílic i un conseqüent augment de la hidrofobicitat. L'anàlisi mitjançant DSC confirma que la realització d'un tractament tèrmic fa augmentar el percentatge de cristal·linitat de les mostres i l'anàlisi per microscòpia electrònica d'escombratge (SEM) revela l'organització esferulítica de l'aspirina en els stents i permet identificar una fragilització del PLLA per degradació del polímer en solució tampó fosfat a 37°C, ja que es visualitzen fractures en els filaments que componen l'stent. L'assaig de citotoxicitat en films mostra que una quantitat excessiva d'AAS implica una viabilitat cel·lular nul·la, mentre que es demostra la biocompatibilitat dels stents carregats amb aspirina quan hi ha agitació del medi. Les imatges obtingudes de l'assaig de trombogenicitat, realitzat amb l'equip Impact-R, mostren una tendència a l'agregació de plaquetes quan els films són sotmesos a un tractament tèrmic i a una degradació hidrolítica de l'AAS. Mitjançant un assaig d'alliberament dut a terme durant un període de dues setmanes en una solució tampó fosfat salí de pH = 7,4, a 37°C i amb agitació, s'ha pogut determinar que els stents de menor superfície específica i que han estat tractats tèrmicament alliberen menor quantitat d'AAS, tot i que el seu alliberament té lloc de forma lineal durant tot l'assaig i estimant un alliberament total d'un mes aproximadament. Per contra, els stents de major superfície específica, presenten una fase d'alliberament ràpid en què alliberen prop del 70% del fàrmac en prop de 48 hores per estabilitzar-se posteriorment. La fabricació d'stents de PLLA carregats de fàrmac per impressió 3D és una bona alternativa a mètodes actuals de fabricació i una estratègia per a obtenir stents adequats a les necessitats del pacient. A més són una bona plataforma obtenint alliberament durant un mes tot i que l'efectivitat del fàrmac disminueix amb el temps i amb el tractament tèrmic efectuat als stents fabricats.

En aquest treball es caracterizaran els efectes del tractament amb plasma a pressio atmosférica, sobre diferents paràmetres de medis líquids d'interes medic / biològic en especial amb respecte a la generació d'especies reactives d'oxigen i de nitrogen, de potencial rellevancia en terapies anti càncer.

Desde hace años se han usado tratamientos con plasma en la industria como métodos de modificación superficial de materiales, muy habitualmente a baja presión. Con el desarrollo reciente de pequeños dispositivos de haz de plasma capaces de trabajar a presión atmosférica, avances científicos recientes apuntan a que es posible dañar e incluso matar células cancerígenas posiblemente debido a la acción de especies reactivas de oxígeno y nitrógeno como principales actuadores, sin afectar a las células sanas. Se forma así un campo en estudio: la medicina de plasmas. En el presente trabajo de final de grado se estudia el efecto que el tratamiento con dos dispositivos de plasma atmosférico genera en el hidrogel de gelatina, que se pretende emplear como vehículo para la liberación de las especies reactivas. Este trabajo se enmarca dentro de un proyecto de investigación con el objetivo último de desarrollar un tratamiento contra el cáncer de huesos lo menos intrusivo posible empleando la combinación de tratamientos de plasma y biomateriales. Para hacerlo, se ha determinado la mejor concentración de gelatina en vistas a la aplicación objeto de estudio y se han tratado muestras de ésta variando parámetros en el tratamiento de plasma, con el fin de establecer las condiciones de tratamiento óptimas. Una vez establecidas, se han realizado ensayos sobre el tiempo de tratamiento, la durabilidad de las especies reactivas en la gelatina y mezclas con hidrogel de alginato. Una de las principales novedades de este trabajo es el de adaptar la metodología de cuantificación de especies reactivas en líquidos para permitir la determinación de dichas especies reactivas generadas con plasma en los hidrogeles de gelatina. Se ha comprobado que se generan especies reactivas (H2O2, NO2- y especies de tiempo de vida corto) en el hidrogel de gelatina cuando éste se trata con plasma atmosférico, con un incremento proporcional en función del tiempo de tratamiento. Se ha comprobado que las especies reactivas H2O2 y NO2- permanecen en el hidrogel de gelatina un tiempo del orden de días y que se generan en mayor concentración en hidrogel de gelatina que en hidrogel de alginato, resultando las mezclas de ambos biopolímeros con concentraciones menores de especies reactivas.
